چکیده

This letter is a report on a comparison between two methods of assessment of dopamine function in a group of abstinent alcoholics, one a neuroendocrine challenge test using a dopaminergic probe, and the other a measurement of platelet monoamine oxidase (MAO) enzyme activity. This enzyme is on the dopamine degradation pathway and has been reported as having diminished activity in alcoholics (Pandey et al, 1988; Faraj et al, 1994). Balldin et al. (1994) reported in this journal evidence that a group of alcoholics with a reduced dopamine D2 receptor function, as explored using a neuroendocrine challenge, relative to healthy controls, possessed normal platelet monoamine oxidase activity. These authors, however, failed to compare the level of platelet MAO activity between alcoholics with and without reduced dopamine function.

Alcohol-dependent patients with neuroendocrine evidence for reduced dopamine d(2) receptor function have decreased platelet monoamine oxidase-B activity.

letter is a report of a re-evaluation of the results obtained in an earlier study of ours (Balldin et al., 1994) of platelet monoamine oxidase (MAO)-B activity in alcoholics with reduced dopamine (DA) D 2 receptor function, as assessed by the growth hormone (GH) response to the D 1 /D 2 agonist apomorphine (APO), which was published in this journal. The reason for the re-evaluation of the resul...

Platelet monoamine oxidase activity in alcoholics with and without a family history of alcoholism.

A number of studies point at platelet monoamine oxidase (MAO) activity being reduced in alcoholics with a family history of drinking, this being a possible vulnerability marker for alcoholism. To test this hypothesis, we examined a group of recently detoxified alcoholics with high (n = 25) and low genetic loading for alcoholism (n = 28) and a group of healthy controls (n = 21). Clinical assessm...

Behavioural and neuroendocrine effects of aqueous extract of Boerhaavia diffusa Linn. in mice using tail suspension and forced swim tests--a preliminary study.

The present study was done to evaluate the effect of aqueous extract of B. diffusa on depression in mice using behavioral models such as tail suspension test (TST) and forced swim test (FST). The extract (50, 100 and 200 mg/kg, po) was administered for 14 successive days to Swiss young albino mice. On 14th day, 60 min after administration, mice were subjected to TST and FST. The administration ...

The urinary MHPG/creatinine ratio and its relationship to platelet monoamine oxidase activity in abstinent alcoholics.

This study was designed to assess the baseline noradrenergic turnover of subgroups of postwithdrawal abstinent alcoholics and healthy controls. The method chosen was an overnight fasting urine sample of the breakdown product of norepinephrine, MHPG, related to urinary creatinine. A comparison was made with platelet monoamine oxidase activity and also within subgroups of the study population. Th...

Adaptive changes in postsynaptic dopamine receptors despite unaltered dopamine dynamics in mice lacking monoamine oxidase B.

Monoamine oxidase (MAO) B is considered a key enzyme in dopamine metabolism. The present studies, conducted in MAO B knockout mice, show that lack of MAO B does not alter extracellular levels of dopamine in striatum. Similarly, the synthesis, storage, uptake, and release of dopamine are also unaltered. However, autoradiography revealed a significant up-regulation of the D2-like dopamine recepto...
